Azithrocin 250 mg is indicated for the treatment of a wide range of bacterial infections caused by susceptible organisms. It is commonly prescribed for infections of the upper respiratory tract, including sinusitis, pharyngitis, and tonsillitis. It is also effective in lower respiratory tract infections such as acute bronchitis and community-acquired pneumonia.

Azithrocin 250 mg is used in the management of otitis media (middle ear infections) and infections of the skin and soft tissues. In addition, it is recommended for the treatment of sexually transmitted infections, including non-gonococcal urethritis and cervicitis caused by Chlamydia trachomatis in both men and women.

Pharmacology

Azithrocin 250 mg belongs to the azalide group within the macrolide class of antibiotics. It works by inhibiting bacterial protein synthesis through binding to the 50S ribosomal subunit of susceptible microorganisms. This action prevents bacterial growth and replication.

Azithromycin demonstrates activity against a broad spectrum of organisms, including gram-positive bacteria such as Staphylococcus aureus and Streptococcus species, and gram-negative bacteria such as Haemophilus influenzae, Moraxella catarrhalis, Neisseria gonorrhoeae, and Escherichia coli. It also shows effectiveness against atypical pathogens like Mycoplasma pneumoniae, Legionella pneumophila, Chlamydia trachomatis, and Borrelia burgdorferi. In vitro studies have shown activity against organisms such as Campylobacter species, Treponema pallidum, Toxoplasma gondii, and Bordetella species.

Dosage

Adults:

• 500 mg once daily for 3 days, or

• 500 mg on day 1 followed by 250 mg once daily on days 2–5

• For sexually transmitted infections caused by Chlamydia trachomatis:

  • 1 g as a single dose, or

  • 500 mg on day 1 followed by 250 mg once daily for the next 2 days

Children (over 6 months):

• 10 mg/kg body weight once daily for 3 days

• 15–25 kg: 200 mg (1 teaspoonful) once daily for 3 days

• 26–35 kg: 300 mg (1½ teaspoonfuls) once daily for 3 days

• 36–45 kg: 400 mg (2 teaspoonfuls) once daily for 3 days

Typhoid Fever:

• 500 mg once daily for 7–10 days

Administration

For suspension preparation:

1. Shake the bottle well to loosen the powder.

2. Add boiled and cooled water up to the marked line on the bottle.

3. Shake thoroughly to ensure complete mixing.

Azithrocin 250 mg should be taken at least one hour before or two hours after meals for optimal absorption.

Drug Interactions

Food and antacids may reduce the absorption of Azithrocin 250 mg. Concomitant use with ergot alkaloids should be avoided due to the risk of ergotism. Caution is advised when used with cyclosporin or digoxin, as macrolides may increase their plasma concentrations. No significant interactions have been observed with warfarin, theophylline, carbamazepine, methylprednisolone, or cimetidine.

Contraindications

Azithrocin 250 mg is contraindicated in patients with known hypersensitivity to azithromycin or other macrolide antibiotics. It should not be used concurrently with ergot derivatives. Patients with severe hepatic impairment should avoid this medication.

Side Effects

Azithrocin 250 mg is generally well tolerated. Common side effects include nausea, vomiting, abdominal discomfort, diarrhea, flatulence, headache, and skin rash. These effects are usually mild and reversible upon discontinuation.

Pregnancy & Lactation

Azithrocin 250 mg is classified as Pregnancy Category B. Animal studies have shown no evidence of fetal harm; however, adequate controlled studies in pregnant women are lacking. The drug should be used during pregnancy only if clearly needed. It is unknown whether azithromycin is excreted in breast milk, so caution is advised when administered to nursing mothers.

Precautions & Warnings

Patients should be monitored for signs of superinfection with non-susceptible organisms, including fungi. No dosage adjustment is required in patients with renal impairment.

Overdose

There is limited information regarding overdose. Symptoms may include severe nausea, vomiting, diarrhea, and reversible hearing loss. Treatment is supportive and may include gastric lavage if necessary.

Storage

Store in a cool, dry place away from heat and light. Keep out of the reach of children.
