A-Clox Injection 500 mg/vial contains Cloxacillin Sodium, a semisynthetic, penicillinase-resistant penicillin antibiotic formulated for parenteral administration. It is used in the treatment of moderate to severe bacterial infections caused by susceptible organisms, particularly penicillinase-producing Staphylococcus species. The injectable form ensures rapid onset of action and reliable therapeutic drug levels, making it suitable for serious infections and hospitalized patients.

A-Clox Injection 500 mg/vial is indicated for the treatment of infections caused by cloxacillin-sensitive organisms, including:
Severe skin and soft tissue infections
Bone and joint infections such as osteomyelitis and septic arthritis
Lower and upper respiratory tract infections
Septicemia and bacteremia
Endocarditis caused by susceptible staphylococci
Post-operative and post-traumatic infections
It is particularly useful when oral therapy is ineffective or not feasible.
Penicillinase-resistant penicillins (β-lactam antibiotics)
Cloxacillin Sodium exerts bactericidal activity by inhibiting bacterial cell wall synthesis. It binds to penicillin-binding proteins, preventing the formation of peptidoglycan, an essential structural component of the bacterial cell wall. This disruption leads to weakening of the cell wall and eventual bacterial lysis.
Cloxacillin is resistant to inactivation by staphylococcal β-lactamase enzymes, allowing it to remain effective against penicillin-resistant strains of Staphylococcus aureus. Following intramuscular or intravenous administration, cloxacillin achieves therapeutic plasma concentrations quickly and is widely distributed throughout body tissues and fluids. The drug is primarily eliminated via renal excretion.
A-Clox Injection inhibits bacterial cell wall synthesis by blocking peptidoglycan cross-linking, resulting in bacterial cell death. Its resistance to β-lactamase enzymes ensures sustained antibacterial efficacy against penicillinase-producing organisms.
Dosage depends on infection severity, patient age, and renal function:
Adults: Typically 500 mg every 6 hours by intramuscular or intravenous injection
Severe infections: Higher or more frequent doses may be required under medical supervision
Pediatric patients: Dose adjusted according to body weight
A-Clox Injection should be administered by a qualified healthcare professional. The duration of therapy should be guided by clinical response and microbiological findings.

Bacteriostatic antibiotics such as tetracyclines may reduce the antibacterial effect of cloxacillin. Probenecid can increase cloxacillin plasma concentrations by decreasing renal excretion. Caution is advised when used with anticoagulants.
A-Clox Injection 500 mg/vial is contraindicated in patients with:
Known hypersensitivity to cloxacillin or other penicillins
History of severe allergic reactions to β-lactam antibiotics
A-Clox Injection is generally well tolerated. Possible side effects include:
Pain, redness, or inflammation at the injection site
Gastrointestinal disturbances
Skin rash or hypersensitivity reactions
Rarely, elevation of liver enzymes or hematological abnormalities
A-Clox Injection may be used during pregnancy and lactation under medical supervision, as cloxacillin is generally considered safe in these populations.
