Sitagliptin works by inhibiting the DPP-4 enzyme, which is responsible for breaking down incretin hormones such as GLP-1 (Glucagon-Like Peptide-1) and GIP (Glucose-Dependent Insulinotropic Polypeptide). By blocking DPP-4:
Incretin hormone levels increase
Insulin secretion from pancreatic beta cells increases in response to meals
Glucagon secretion from alpha cells decreases
Hepatic glucose production is reduced
Because its action is glucose-dependent, Sitagliptin primarily works when blood sugar levels are elevated, minimizing the risk of excessive hypoglycemia.
Janvia Tablet 50 mg is indicated for:
Management of Type 2 Diabetes Mellitus in adults
Monotherapy when diet and exercise are insufficient
Combination therapy with Metformin, sulfonylureas, insulin, or other antidiabetic agents
Patients requiring improved glycemic control with a low risk of hypoglycemia
It is not indicated for Type 1 diabetes or diabetic ketoacidosis.
The usual recommended dose is 50 mg once daily, or as prescribed by a healthcare professional. Dose adjustment may be required in patients with renal impairment. The tablet can be taken with or without food and should be swallowed whole with water.
