Losucon M Tablet 1 mg+500 mg is a fixed-dose combination of Glimepiride and Metformin, two well-established antidiabetic agents with complementary mechanisms of action. It is used in the management of Type II diabetes mellitus (non-insulin dependent diabetes mellitus, NIDDM) in patients whose blood glucose levels are not adequately controlled with diet, exercise, or monotherapy alone. By combining these two agents, Losucon M helps achieve better and more sustained glycaemic control.

Losucon M Tablet is indicated for:
The treatment of Type II diabetes mellitus as an adjunct to diet and exercise.
Patients inadequately controlled on metformin or sulfonylurea monotherapy alone.
Patients already receiving Glimepiride and Metformin as separate tablets who require combination therapy for improved glycaemic control.
Sulfonylurea and Biguanide combination
Losucon M Tablet works through dual mechanisms. Glimepiride, a sulfonylurea, stimulates insulin release from functioning pancreatic beta cells and enhances the sensitivity of these cells to glucose. It also improves peripheral insulin sensitivity, aiding glucose uptake by muscles and adipose tissue.
Metformin, a biguanide, primarily reduces hepatic glucose production, decreases intestinal absorption of glucose, and improves insulin sensitivity at peripheral tissues. Unlike sulfonylureas, metformin does not stimulate insulin secretion and therefore does not usually cause hypoglycaemia when used alone. Together, these mechanisms help lower both fasting and postprandial blood glucose levels effectively.
The dosage of Losucon M Tablet should be individualized based on the patient’s blood glucose levels and previous antidiabetic therapy.
The tablet is usually taken once or twice daily with meals to reduce gastrointestinal side effects.
Dose adjustments should be made gradually under medical supervision, based on regular monitoring of blood glucose levels.
Patients should adhere strictly to dietary advice and exercise regimens while taking this medication.
Losucon M Tablet may interact with several medicines. Drugs that enhance the hypoglycaemic effect include insulin, other oral antidiabetic agents, ACE inhibitors, salicylates, sulfonamide antibiotics, and fluconazole. Corticosteroids, diuretics, thyroid hormones, and sympathomimetic agents may reduce its glucose-lowering effect. Alcohol intake should be avoided, as it may increase the risk of hypoglycaemia and lactic acidosis associated with metformin.

Losucon M Tablet is contraindicated in patients with:
Type I diabetes mellitus
Diabetic ketoacidosis or diabetic coma
Severe renal or hepatic impairment
Conditions associated with hypoxia
Hypersensitivity to Glimepiride, Metformin, or other sulfonylureas
Common side effects may include hypoglycaemia, nausea, vomiting, diarrhoea, abdominal discomfort, metallic taste, dizziness, headache, and fatigue. Rarely, metformin may be associated with lactic acidosis, particularly in patients with renal impairment.
Losucon M Tablet is not recommended during pregnancy. Patients planning pregnancy or who become pregnant should be switched to insulin therapy. It should also be avoided during breastfeeding, as the components may pass into breast milk and affect the infant.
