Lubistone 8 mcg is a locally-acting chloride channel activator designed to relieve constipation by increasing intestinal fluid secretion and motility. It is indicated for adults with Irritable Bowel Syndrome with Constipation (IBS-C), Chronic Idiopathic Constipation (CIC), and Opioid-Induced Constipation (OIC) in patients with chronic non-cancer pain. By enhancing fluid secretion and promoting smooth bowel movements, Lubistone 8 mcg helps alleviate discomfort, bloating, and abdominal pressure associated with constipation.

Lubistone 8 mcg is prescribed for:
Irritable Bowel Syndrome with Constipation (IBS-C): Improves bowel movement frequency and stool consistency
Chronic Idiopathic Constipation (CIC): Provides relief from persistent constipation in adults
Opioid-Induced Constipation (OIC): Helps counteract constipation caused by opioid therapy in chronic, non-cancer pain patients
Lubistone 8 mcg works by activating ClC-2 chloride channels on the apical membrane of the intestinal epithelium. Activation of these channels leads to an efflux of chloride ions into the intestinal lumen, followed by sodium and water, increasing intestinal fluid secretion. This mechanism:
Enhances intestinal motility and eases stool passage
Restores mucosal barrier function by supporting tight junction proteins
Provides targeted action in the gut with minimal systemic absorption
IBS-C: 8 mcg twice daily with food and water. In severe hepatic impairment, start at 8 mcg once daily and increase if tolerated.
CIC & OIC: 24 mcg twice daily with food and water. In moderate hepatic impairment, start at 16 mcg twice daily; in severe impairment, start at 8 mcg twice daily.
Take orally with food and water to reduce nausea and improve tolerance.
Efficacy may be reduced in patients using diphenylheptane opioids.
Known or suspected mechanical gastrointestinal obstruction
Common adverse reactions include:
Diarrhea
Abdominal bloating or pressure
Nausea
Stomach pain or swelling
Dyspnea
Monitor for nausea; taking Lubistone with food may help
Avoid in patients with severe diarrhea
Rule out mechanical bowel obstruction before starting therapy
Symptoms may include: nausea, diarrhea, vomiting, dizziness, abdominal pain, flushing, dyspnea, pallor, chest discomfort, dry mouth, syncope, and hyperhidrosis. Supportive care is recommended.

Lubistone 8 mcg increases intestinal fluid secretion and motility through selective activation of ClC-2 chloride channels, aiding stool passage and improving bowel regularity. It acts primarily at the apical portion of the gastrointestinal epithelium, producing minimal systemic effects.
Pregnancy Category C: Use only if the potential benefit justifies the risk
Excretion in human milk is unknown; caution is advised, and breastfed infants should be monitored for diarrhea
Store between 15–30°C in a cool, dry place, protected from light. Keep out of reach of children.
