Protolan 30 mg is a proton pump inhibitor (PPI) formulated for the effective management of acid-related gastrointestinal disorders. It contains a substituted benzimidazole compound that works by profoundly reducing gastric acid secretion. Protolan 30 mg is widely used in the short-term and maintenance treatment of peptic ulcer disease, gastroesophageal reflux disease (GERD), and other conditions associated with excessive gastric acid production.

Protolan 30 mg acts by selectively inhibiting the H⁺/K⁺ ATPase enzyme system located in the gastric parietal cells. This enzyme represents the final step in gastric acid production. By blocking this pathway, Protolan effectively suppresses both basal and stimulated acid secretion, leading to reduced gastric acidity and promotion of mucosal healing. Due to its targeted mechanism, it provides sustained acid suppression with once-daily dosing in most patients.
Protolan 30 mg is indicated for the treatment and management of the following conditions:
Short-term treatment of active duodenal ulcer
Maintenance of healed duodenal ulcers
Short-term treatment of active benign gastric ulcers
Short-term treatment of active erosive esophagitis
Maintenance of healing of erosive esophagitis
Gastroesophageal reflux disease (GERD)
Acid-related dyspepsia
Pathological hypersecretory conditions, including Zollinger–Ellison syndrome
Helicobacter pylori eradication (as part of combination therapy) to reduce the risk of duodenal ulcer recurrence
Dosage should be individualized according to the condition being treated and patient response:
Benign gastric ulcer: 30 mg once daily in the morning for 8 weeks
Duodenal ulcer: 30 mg once daily for 4 weeks; maintenance dose 15 mg
NSAID-associated ulcers: 15–30 mg daily for 4 weeks, extendable to 8 weeks if required
GERD: 30 mg daily for 4 weeks; may be extended to 8 weeks; maintenance 15–30 mg daily
Zollinger–Ellison syndrome: Initially 60 mg once daily, adjusted according to response; doses above 120 mg should be given in divided doses
Acid-related dyspepsia: 15–30 mg daily for 2–4 weeks
Protolan 30 mg should preferably be taken in the morning before food.
Protolan 30 mg is a selective inhibitor of the cytochrome P-450 monooxygenase system. Although it may affect hepatic clearance of certain drugs, no clinically significant interactions have been widely reported. Reduced gastric acidity may influence vitamin B12 absorption and alter gastric bacterial colonization during long-term use.

Protolan 30 mg is contraindicated in patients with known hypersensitivity to any component of the formulation.
Common side effects include headache and dose-dependent diarrhea. Long-term use may lead to increased serum gastrin levels. In patients with Helicobacter pylori infection, prolonged therapy may accelerate the development of atrophic gastritis. Rare but serious effects related to prolonged acid suppression have been reported.
Protolan 30 mg should be avoided during pregnancy and lactation unless clearly indicated and prescribed by a physician.
Gastric malignancy should be excluded prior to therapy. Caution is advised in patients with hepatic impairment and during prolonged use.
Neonates: Not recommended due to lack of data
Children: Clinical experience exists in patients aged 13 years and above
Elderly: No dosage adjustment is generally required
Store at temperatures not exceeding 25°C. Protect from moisture and keep out of reach of children.
