Herpes Simplex & Varicella-Zoster Virus Infections
Valovir 1 gm is indicated for:
Herpes Zoster (Shingles): Rapidly reduces viral replication and accelerates healing
Genital Herpes:
Initial episode: Treats infection effectively
Recurrent episodes: Shortens symptom duration
Suppressive therapy: Reduces frequency of recurrences in immunocompetent and HIV-infected patients
Cold Sores (Herpes Labialis): Reduces healing time and severity of outbreaks
Valovir 1 gm is converted in the body to acyclovir, which selectively inhibits viral DNA polymerase, blocking viral replication. This limits the spread of the virus to healthy cells and reduces the severity and duration of symptoms.
Key pharmacokinetic points:
Rapidly absorbed from the gastrointestinal tract
Converted to active acyclovir in the liver and intestines
Excreted primarily via the kidneys
Peak plasma levels are achieved within 1–2 hours of oral administration
Adults:
Cold Sores: 2 grams every 12 hours for 1 day
Genital Herpes:
Initial episode: 1 gram twice daily for 10 days
Recurrent episode: 500 mg twice daily for 3 days
Suppressive therapy:
Immunocompetent: 1 gram once daily or 500 mg once daily for < 9 recurrences/year
HIV-infected: 500 mg twice daily
Herpes Zoster: 1 gram three times daily for 7 days
Pediatric Patients:
Cold sores (>12 years): 2 grams every 12 hours for 1 day
Chickenpox (2–18 years): 20 mg/kg three times daily for 5 days (max 1 gram three times daily)
Administration Tips:
Take with or without food
Maintain adequate hydration to prevent kidney complications
No dose adjustments are required when coadministered with digoxin, antacids, thiazide diuretics, cimetidine, or probenecid in patients with normal renal function
Known hypersensitivity to Valovir, acyclovir, or any excipient in the formulation
Common side effects include:
Nausea (15%)

Abdominal pain (3%)
Most side effects are mild and transient.
Dose adjustment is recommended for renal impairment
Exercise caution in elderly patients or those receiving nephrotoxic drugs

Pregnancy Category B: No evidence of teratogenicity in animal studies
