Abemaciclib works by inhibiting cyclin-dependent kinases 4 and 6 (CDK4/6), which are enzymes responsible for controlling cell division. These kinases normally help cancer cells progress through the cell cycle from the G1 phase to the S phase, allowing them to multiply rapidly.
By blocking CDK4/6 activity, abemaciclib prevents phosphorylation of the retinoblastoma protein and stops cancer cells from dividing and growing. As a result, tumor growth slows and cancer cells may eventually die.
The tablet is taken orally with or without food.
It should be swallowed whole with water and not crushed or chewed.
