Oxazolidinone Antibiotic
Linezolid, the active ingredient in Linzolid Tablet, works by inhibiting bacterial protein synthesis. It binds to the 23S portion of the 50S ribosomal subunit, preventing the formation of the 70S initiation complex, a critical step in bacterial translation. This action is bacteriostatic against enterococci and staphylococci and bactericidal against most streptococci.
Linzolid demonstrates excellent oral bioavailability, with absorption unaffected by food. It distributes widely into tissues, including the lungs, skin, and soft tissue, making it suitable for a range of systemic infections. The drug is metabolized minimally in the liver, and the majority is excreted in urine as metabolites, with a half-life of approximately 5–7 hours.
Linzolid Tablet 600 mg is indicated for the treatment of:
Complicated skin and soft tissue infections caused by susceptible Gram-positive bacteria
Nosocomial and community-acquired pneumonia
Infections caused by MRSA and VRE
Other serious Gram-positive bacterial infections when alternative treatments are ineffective or inappropriate
Adults: The usual dose is 600 mg every 12 hours for 10–14 days, depending on the severity and site of infection.
Renal or hepatic impairment: No dosage adjustment is generally required, but monitoring is advised in severe dysfunction.
Administration: Tablets should be swallowed whole with or without food. Consistency in timing ensures optimal blood levels.
Hypersensitivity to Linezolid or any component of the formulation
Concomitant use with monoamine oxidase inhibitors (MAOIs) due to risk of serotonin syndrome
History of linezolid-induced myelosuppression
Linzolid is generally well tolerated, but some adverse effects may occur:
Hematologic: Thrombocytopenia, anemia, leukopenia, and pancytopenia, particularly with therapy beyond 2 weeks
Gastrointestinal: Nausea, vomiting, diarrhea, and taste disturbances
Neurological: Headache, dizziness, peripheral neuropathy (rare with prolonged use)
Other: Lactic acidosis and optic neuropathy in prolonged therapy
Regular complete blood count monitoring is recommended during therapy, especially for courses longer than 2 weeks
Use caution in patients with renal or hepatic impairment
Avoid tyramine-rich foods and certain medications to reduce risk of hypertensive reactions or serotonin syndrome
Pregnancy & Lactation: Linezolid should be used only if clearly needed; excretion in breast milk occurs, so breastfeeding is not recommended during treatment
Store in a cool, dry place below 30°C, protected from light and moisture
