Amoxicillin is a broad-spectrum, bactericidal penicillin antibiotic that works by inhibiting the biosynthesis of bacterial cell walls, leading to bacterial cell death. It is stable in gastric acid and is rapidly absorbed after oral administration. Peak plasma concentrations are reached within 1 to 2 hours. Amoxicillin is widely distributed throughout body tissues and fluids, allowing effective treatment of infections at various sites.

Indications

Tycil 125 mg/5 ml Suspension is indicated for the treatment of infections caused by susceptible organisms, including:

• Ear, Nose, and Throat Infections: Otitis media, sinusitis, tonsillitis, pharyngitis, laryngitis

• Lower Respiratory Tract Infections: Pneumonia, acute and chronic bronchitis, lung abscess, empyema, bronchiectasis

• Skin and Soft Tissue Infections: Cellulitis, carbuncles, furunculosis, infected wounds, abscesses

• Genito-Urinary Tract Infections: Pyelonephritis, cystitis, urethritis

• Venereal Disease: Acute uncomplicated gonorrhoea

• Dental Infections: Short-term treatment of dental abscess

• Gastrointestinal Indication: In combination with clarithromycin and lansoprazole as part of triple therapy for Helicobacter pylori infection and duodenal ulcer disease, and to reduce the risk of duodenal ulcer recurrence

Dosage and Administration

Dosage depends on patient age, body weight, severity of infection, and clinical response.

• Adults:

  • Usual dose: 250 mg three times daily

  • Severe infections: Up to 500 mg three times daily

• Children (up to 10 years):

  • Usual dose: 125 mg three times daily

  • Severe infections: Up to 250 mg three times daily

• Special Dosage Recommendations:

  • Severe or recurrent purulent respiratory infections: 3 g every 12 hours

  • Otitis media: Adults 1 g three times daily; children 40 mg/kg/day in divided doses (maximum 3 g daily)

  • Pneumonia: 500–1000 mg three times daily

  • Dental abscess and urinary tract infections: 3 g, repeated after 10–12 hours

  • Gonorrhoea: Single dose of 2–3 g with probenecid 1 g

Dose reduction may be necessary in patients with renal impairment.

The reconstituted suspension may be mixed with water, milk, or fruit juice and should be taken immediately after preparation.

Drug Interactions

• Concurrent use with probenecid may increase and prolong blood levels of Amoxicillin

• May reduce the effectiveness of combined oral estrogen/progesterone contraceptives due to altered gut flora

Contraindications

Tycil 125 mg/5 ml is contraindicated in patients with known hypersensitivity to penicillins.

Side Effects

Side effects are generally mild and transient and may include diarrhoea, indigestion, nausea, or skin rash. Rarely, pseudomembranous colitis has been reported.

Pregnancy and Lactation

Amoxicillin is classified as US FDA Pregnancy Category B. It should be used during pregnancy only if clearly needed. As Amoxicillin is excreted in breast milk, caution should be exercised when administered to lactating mothers.

Precautions and Warnings

Prolonged use may lead to superinfections with resistant bacteria or fungi. If superinfection occurs, Amoxicillin should be discontinued and appropriate therapy initiated.

Store in a dry place away from light and heat. Keep out of the reach of children.
