Amdopril Tablet 5 mg+5 mg is a combination therapy containing perindopril arginine 5 mg and amlodipine 5 mg, designed for the treatment of hypertension and cardiovascular risk management. By combining an ACE inhibitor (perindopril) with a calcium channel blocker (amlodipine), Amdopril provides a complementary dual mechanism to lower blood pressure, reduce strain on the heart, and protect cardiovascular organs. This fixed-dose combination is particularly beneficial for patients whose blood pressure is not adequately controlled with monotherapy.

Antihypertensive / ACE Inhibitor + Calcium Channel Blocker
Amdopril Tablet 5 mg+5 mg is indicated for:
Hypertension: Management of high blood pressure in adults to reduce the risk of stroke, myocardial infarction, and other cardiovascular complications.
Cardiovascular Risk Reduction: In patients with additional risk factors, such as diabetes or left ventricular hypertrophy, the combination helps prevent end-organ damage.
Patients Not Controlled on Monotherapy: Suitable for patients requiring dual therapy for effective blood pressure control.
Perindopril Arginine:
Perindopril is an angiotensin-converting enzyme (ACE) inhibitor. It blocks the conversion of angiotensin I to angiotensin II, a potent vasoconstrictor. This leads to vasodilation, reduced peripheral vascular resistance, and lower blood pressure. By reducing the effects of angiotensin II, perindopril also decreases aldosterone secretion, which reduces sodium and water retention, further contributing to blood pressure control and cardiac protection.
Amlodipine:
Amlodipine is a dihydropyridine calcium channel blocker that inhibits calcium ion influx in vascular smooth muscle cells. This causes vasodilation of peripheral arteries, decreases systemic vascular resistance, and improves coronary blood flow.
Combined Effect:
The combination of perindopril and amlodipine offers synergistic antihypertensive effects, as the ACE inhibitor prevents compensatory vasoconstriction and sodium retention caused by amlodipine-induced vasodilation. Together, they provide more effective and sustained blood pressure reduction than either agent alone.
Typical Adult Dose: One tablet once daily, preferably at the same time each day.
Administration: Tablets should be swallowed whole with water and can be taken with or without food.
Dose Adjustment: The dose may be adjusted based on patient response and tolerability. Renal function should be monitored, especially in patients with pre-existing kidney disease.
Lifestyle modifications, including dietary salt restriction, exercise, and weight management, are recommended to enhance therapeutic outcomes.
Amdopril Tablet 5 mg+5 mg should not be used in patients with:
Hypersensitivity to perindopril, amlodipine, or any excipients
History of angioedema related to ACE inhibitors
Severe hypotension or cardiogenic shock
Severe aortic stenosis or unstable heart failure
Pregnancy and lactation (unless clearly necessary)
Common side effects include:
Dizziness, headache, fatigue
Peripheral edema, flushing
Cough (associated with ACE inhibitor)
Palpitations
Rare but serious adverse events may include angioedema, hypotension, electrolyte imbalance, renal impairment, or liver function abnormalities.
Monitor blood pressure, kidney function, and electrolytes regularly.
Caution in patients with heart failure, renal impairment, or diabetes.
Avoid abrupt discontinuation to prevent rebound hypertension.
Perindopril is contraindicated during pregnancy, especially in the second and third trimesters, due to risk of fetal toxicity. Caution is advised during breastfeeding. Amlodipine should also be used during pregnancy only if benefits outweigh risks.
Store below 30°C in a dry place, protected from light and moisture. Keep out of reach of children.
