Eltromin Tablet 50 mg contains Eltrombopag Olamine, a thrombopoietin receptor agonist used to treat low platelet counts (thrombocytopenia) in certain conditions. It is particularly effective for patients with chronic immune thrombocytopenia (ITP), thrombocytopenia associated with chronic hepatitis C, and aplastic anemia. By stimulating platelet production, Eltromin helps reduce the risk of bleeding complications and improves patient quality of life.

Eltrombopag is an oral medication, making it convenient compared to injectable treatments, and has a selective action on the c-Mpl receptor, which regulates platelet production.

Pharmacology

Eltromin’s active ingredient, Eltrombopag Olamine, works by binding to the thrombopoietin (TPO) receptor on megakaryocyte precursors in the bone marrow. This activation leads to:

  1. Increased platelet production – Promotes differentiation and proliferation of megakaryocytes, which are platelet-producing cells.

  2. Enhanced platelet counts in peripheral blood – Reduces the risk of bleeding in patients with chronic thrombocytopenia.

  3. Selective receptor activation – Does not stimulate other hematopoietic lineages significantly, minimizing off-target effects.

Eltrombopag is absorbed orally, with peak plasma levels reached within 2–6 hours. It undergoes hepatic metabolism, mainly via glucuronidation and oxidation, and is excreted predominantly in feces. The drug has a half-life of approximately 21–32 hours, allowing for once-daily dosing.


Indications

Eltromin Tablet 50 mg is indicated for:

  • Chronic Immune Thrombocytopenia (ITP) – In adults and children who have had an inadequate response to corticosteroids, immunoglobulins, or splenectomy.

  • Thrombocytopenia in Chronic Hepatitis C – To increase platelet counts in patients who require interferon-based antiviral therapy.

  • Severe Aplastic Anemia – In combination with immunosuppressive therapy for patients unresponsive to other treatments.

By increasing platelet levels, Eltromin reduces the risk of spontaneous or procedure-related bleeding.


Dosage and Administration

  • Adults with ITP: Initial dose is usually 50 mg once daily; may be adjusted based on platelet response.

  • Children (1–17 years) with ITP: Dosing varies according to body weight and clinical response.

  • Hepatitis C patients: Typically 25–50 mg once daily, adjusted according to platelet counts during antiviral therapy.

  • Tablets should be taken on an empty stomach, at least 1 hour before or 2 hours after meals containing calcium, dairy, or other polyvalent cations, as these can interfere with absorption.

  • Dose adjustments should be made based on platelet counts, and therapy should be paused if platelet levels exceed safe thresholds.


Contraindications

  • Hypersensitivity to Eltrombopag Olamine or any excipient

  • Severe hepatic impairment without monitoring


Side Effects

Common side effects include:

  • Headache

  • Nausea and vomiting

  • Fatigue

  • Diarrhea or abdominal pain

Serious adverse effects (rare) may include:

  • Thrombosis (blood clots)

  • Hepatic dysfunction (elevated liver enzymes)

Mode of Action
